MBP is one of the principal components of the myelin membrane or sheath. Autoantibodies against MBP can lead to demyelination of nerves and contribute to the formation of gliotic scar tissue. Several studies suggest that the formation of anti MBP antibodies is a common sequel to central nervous system injury.

MBP antibodies are found in MS patients as well as in patients with other inflammatory neurological diseases and non-inflammatory CNS diseases. In MS, MBP antibodies are not established in the sera of affected patients until later as these antibodies tend to accumulate with time.
