KOH (Potassium Hydroxide) Test

Description

KOH mount is one of the principal methods for investigating fungal
infections. Potassium hydroxide (KOH) preparation, also known as the KOH
test is used for the rapid detection of fungal elements in different clinical
specimens. It is an initial screening tool that detects fungal elements
present but does not identify the fungal species. It is commonly used to
differentiate dermatophytes and Candida albicans symptoms from other
nonfungal skin disorders. KOH is a strong alkali, the clinical sample such
as skin, hair, nails or sputum is mixed with KOH preparation, it digests and
clears the tissues making fungal elements more observable during direct
microscopic examination.

Indications

KOH preparation is recommended in suspected conditions of Aspergillus, Dermatophytes (ringworm fungi), Blastomyces dermatitidis and Mucormycosis infections but not exclusively. KOH test can be performed in a wide range of clinical specimens as sputum, skin scrapings, nails, hair, pus, sputum and exudates from infected lesions or tissue. Finding fungal elements in KOH preparation such as hyphae (septated/aseptated), pseudohyphae, spores or yeast cells (spherical, oval or budding) indicate a Positive result.

Collect samples in a sterile, leak-proof container. Specimens should be transported to the lab as soon as possible. All request should have complete and sufficient clinical data.
